摘要:以健康刺参肠道的益生菌——枯草芽孢杆菌(Bacillus subtilis)为研究对象,以豆粕、葡萄糖和氯化钠为基础发酵培养基,通过单因素和正交试验,确定该菌株适用于工业生产的最佳发酵培养基配方;通过单因素试验,确定最佳发酵条件;同时在生物反应器中进行扩大培养试验,得到其最佳种龄和发酵时间。试验确立该菌株的最佳培养基配方为:氯化钠10 g/L,葡萄糖2.5 g/L,豆粕20 g/L;最佳发酵条件为:培养温度 28 ℃,初始pH 7.5,转速170 r/min,菌种接种量5%。以最佳发酵条件和培养基,在10 L生物反应器中进行扩大培养试验,得到最佳种龄为24 h、最佳发酵时间为32 h,菌体细胞浓度最高达到8.5×109 cfu/mL,芽孢生成率达到91%。研究结果为刺参微生态制剂的工业化生产提供基础数据,为海水养殖专用微生态制剂的开发提供参考。

Optimization of fermentation medium and process for a probiotic strain Bacillus subtilis isolated from sea cucumber Apostichopus japonicas

Abstract: The culture medium composition and fermentation condition were studied on a strain Bacillus subtilis (CCTCC M2010316), which has been isolated from healthy sea cucumber Apostichopus japonicus. By univariate tests and orthogonal experiment, the optimal medium composition was as follows: glucose 2.5 g/L, soybean meal 20 g/L, sodium chloride 10 g/L, and the best fermentation conditions was temperature at 28 ℃, initial pH value 7.5, shaker revolution 170 r/min, bacterial inoculum 5%. The pilot scale cultivation revealed that the best inoculum age and fermentation time was 24 h and 32 h, respectively. The highest bacterial concentration was 8.5×109 cfu/mL, spores formation rate reached 91%. This paper would provide basic data for large-scale production of sea cucumber probiotics.
